摘 要:目的 优化利用角叉菜胶诱导小鼠尾部血栓模型形成的条件,提高模型制备的成功率。方法 分别采用腹腔注射与皮下注射的方式给予小鼠不同浓度的角叉菜胶(0、20、50、80、120、150mg/kg bw),对比后得到合适的给药方式和剂量。最后经生理切片证实小鼠尾部血栓制备成功。结果 在注射不同浓度角叉菜胶24h后,腹腔注射方式的各组剂量的成功率都大于50%,而皮下注射方式中,成功率均未超过50%。当角叉菜胶浓度为20mg/kg•bw时,以腹腔注射方式造模成功率就已经超过80%。通过制备生理切片进一步观察,小鼠尾部病变为血栓症状,说明制备血栓模型成功。结论 腹腔注射方式优于皮下注射方式,温度为18±2℃的环境中,20mg/kg•bw浓度的角叉菜胶,制备小鼠血栓模型成功率较高。

Abstract:Objective To enhance the success rates of the mouse tail-thrombus model,the condition of the thrombosis model making was optimized. Methods Carrageenan was injected subcutaneously or intraperitoneally by different density (0, 20, 50, 80, 120, 150mg/kg bw ), and the number of dark tails were counted and the lengths of the dark tails’ (and the whole tails) were measured. At last the paraffinsection on the dark tail was made to verify the thrombosis. Results 24 hours after the injection of different concentration of carrageenan , the success rates of intraperitoneal injection were all larger than 50% in each group; but the success rates of the subcutaneous injection were all less than 50%. When the carrageenan is 20mg/kg·bw, the success rates of intraperitoneal injection were larger than 80%. Through observing the tissue slices, we confirmed that the appearance of dark tail was because of thrombosis in the mouse tail. Conclusions The experiment would be relatively successful while the dose of carrageenan is 20mg/kg•bw and in 18±2℃ambient environment.
