摘要:本文采用热水浸提法、复合酶提取法、超声提取法、超声-微波协同法提取灰树花多糖,通过单因素、正交试验,寻求最佳提取工艺,并对各种方法进行比较。结果表明,热水浸提法最佳工艺为:液料比30ml/g、温度90℃、时间2.5h、pH7.0时,有最佳得率为6.23%;酶解提取法最佳工艺为:液料比20ml/g、时间3h、温度37℃、酶浓度3%、pH7.5时,有最佳得率为8.91%;超声提取法最佳工艺为:液料比30ml/g、时间45min、温度50、37℃、功率500W、pH6.5时,有最佳得率为6.42%;超声-微波协同提取法最佳工艺为:液料比30ml/g、时间15min、功率500W、pH6.5时,有最佳得率为7.17%。其中酶解法多糖得率最高为8.91%。

Abstract:The article aked the comparation of several extraction methods of Grifola frondosa polysaccharide,which included the extraction method of hot water,multi-enzymatic method,ultrasonic wave method, and ultrasonic wave- microwave equip-ment method.By single factor and orthogonal tests,seeking the best extraction process, and compareing various methods. The study showed that the hot water optimal hydrolytic conditions were as follows: liquid ratio of 30, temperature of 90℃, time of 2 h, and pH of 7.0. Under these conditions, the yield was 6.23%. The multi-enzymatic method optimal hydrolytic conditions were as follows: liquid ratio of 20, time of 3 h,temperature of 37℃, the enzyme concentration of 3%, pH of 7.5. Under these conditions, the yield was up to 8.91%.The ultrasonic wave method optimal hydrolytic conditions were as follows: liquid ratio of 30, time of 45 min,temperature of 37℃,power of 500W,and pH of 6.5. Under these conditions, the the yield was 6.42%. The ultrasonic wave- microwave equip-ment method optimal hydrolytic conditions were as follows: liquid ratio of 30,time of 15 min,power of 500W,and pH of 6.5,Under these conditions, the yield was 7.17%.The results showed that multi-enzymatic extraction procedure had the highest extraction rate,the yield was 8.91%.
